I have made a mess of prioritizing my library. How do I undo it and start over?

I believe you will need to do this one resource at a time - see http://wiki.logos.com/Prioritizing#Removing_resources_in_the_Prioritize_panel for details.
Alternatively you can drag resources around in the Prioritization tab to produce your required result
