Highlighting while in Reading View Mode

When a book is in Show in Reading View Mode, is there a way to also have the Highlighting available? In other words, how would one use Highlighting when in Reading View Mode?

Probably like this: Selecting text and using the keyboard shortcut I have assigned to the respective highlight (Y for yellow, G for green, O for Orange...)
