The release notes for Logos 4.5 state: Moved “New palette” function to toolbar. I can't seem to find this palette.
If you open the Highlight panel, you'll find it in the toolbar of that panel (not your main Logos toolbar).
They meant the toolbar on the Highlighting tab (which you bring up using Tools > Highlighting):

On a slightly different vein, is it possible to hightlight parts of a note file [for emphasis in your own note].

It's not possible to highlight notes files using the highlighting panel, but it is possible to change the background colour of parts of a notes file, as Jonathan has illustrated above.
