Can someone show me how please? Thank you.

That's not quite right. You need a + in there before the - . So the correct URL is
https://vyrso.com/products/search?q=+-GenreGroup%3AFiction
You need a + in there before the -
As far as I can tell a + is not strictly necessary at the beginning of the search query, e.g.:
https://vyrso.com/products/search?q=-GenreGroup%3AFiction+-GenreGroup%3AYoung*+-GenreGroup%3ACooking+-GenreGroup%3AHealth*
(this url will also exclude young adult fiction, cooking, and health books).
Edit:
After reading Rosie's post in another thread I realized the url can be made shorter:
https://vyrso.com/products/search?q=-GenreGroup%3A*Fiction+-GenreGroup%3ACooking+-GenreGroup%3AHealth*
The following post shows how one can avoid fiction right in the Vyrso search box (without typing a full URL):

Hi, I would like help pulling out my wife's Christian Fiction books please....any suggestions?
This way she knows what we have in Vyrso for her to enjoy. I would please like to Christian Fiction books filter for her from what I have bought already and maybe for her to see what else in there to buy too. Can you help please?
Thanks.

It would be best to create a new thread... especially since you are asking something quite different than the OP... [;)]
Are you wanting this for a mobile device or desktop? Which version of which app? FYI - The iOS version of the VYRSO app has added a filter to show only edition:e resources (Vyrso books). If you don't buy many Vyrso books other than novels, this will work well. Otherwise, you might have to use community tags or create your own.
One method: Add a tag to the books you want her to see and use the syntax mytag:tagname
